  •        核仁小RNA (snoRNA)是一类中等长度的非编码小RNA,其长度60-400nt不等,与核仁核糖核蛋白结合形成snoRNPs复合物发挥功能[1]。在脊椎动物中,核仁小RNA的编码基因主要存在于蛋白编码基因的内含子区,经转录加工处理形成成熟的核仁小RNA分子[2]。snoRNA参与多种生物学过程,比如rRNA的加工处理,RNA剪接和翻译,以及氧化应激反应的调控[3]。snoRNA主要通过snoRNPs复合物来发挥作用。通过snoRNA碱基互补识别靶基因特定位点,snoRNPs复合物催化这些位点的2’-O甲基化或假尿嘧啶化反应。根据snoRNA结构与功能的不同,可以将snoRNA分为两大类:负责2’-O甲基化的box C/D snoRNA和负责假尿嘧啶化的box H/ACA snoRNA [1]。另外,还存在一类比较特殊的snoRNA- scaRNAs (它们具有类似box C、box D或者box H、box ACA结构,特异表达于small Cajal body) [4]。snoRNA可以产生更小片段的非编码RNA-sdRNA。sdRNA与miRNA类似,能够通过调节mRNA的稳定性来发挥作用 [5]。

           多项研究表明,snoRNA在肿瘤中异常表达,并在肿瘤的转移过程中扮演着重要角色 [6-7]。在乳腺癌和前列腺癌中,SNORD50A发生了2个碱基的缺失突变,并且其拷贝数明显降低,因此被认为是抑癌基因[8]。有趣的是,H/ACA box snoRNA SNORA42在肺癌中作为癌基因存在,抑制SNORA42的表达具有明显的抗癌作[10]用。C/D box snoRNAs的表达水平常伴随肿瘤的发生而增加 [11]。有一些C/D box snoRNAs可以作为疾病的分子标志物,比如SNORD44和SNORD43与乳腺癌的不良预后相关[9]。snoRNA在神经退行性疾病的发生发展过程也发挥着关键作用。snoRNAs在血液、痰液以及尿液中稳定存在,使其作为疾病分子诊断标志物方面具有巨大的前景。目前,snoRNA已经成为疾病领域特别是癌症领域里的研究热点。
           nrStar™ Human snoRNA PCR芯片包括359条snoRNA、7条snoRNA靶向snRNA以及4条snoRNP复合物成员mRNA,而nrStar™ Mouse snoRNA PCR芯片分别包含370个snoRNA。该PCR芯片覆盖了所有PCR技术可区分的snoRNA,能够快速方便的检测出组织、细胞以及血液中的snoRNA的表达情况。它还可应用于疾病分子标志物的高通量筛选和验证。
           为了确保数据质量,nrStar™ Human snoRNA PCR芯片包含3个针对人snoRNA的内参基因和2个针对人snoRNP复合物成员mRNA的内参基因,而nrStar™ Mouse snoRNA PCR芯片包含8个针对小鼠snoRNA的内参基因用于数据的均一化处理。此芯片还使用RNA Spike-in,PCC和GDC对照组来检测cDNA合成效率,PCR扩增效率和基因组DNA污染。

           为保证nrStar™ snoRNA PCR Array(H/M)的实验效果,建议配套使用Arraystar rtStar™ First-Strand cDNA Synthesis Kit (AS-FS-001)
